Maurice Elvey was a good example of a prolific director, having made over 100 films, but as his career stretches back into the silent era I imagine most of them are only shorts: http://www.imdb.com/name/nm0002061/
In the realm of Hollywood feature films, William Beaudine is credited with the most, at 182. Richard Thorpe, who trails by 3 with 179, directed more A and B list films, less serials and explotation films than Beaudine, so he would have to be considered as well.
Spanish director Jesus Franco also has over 180 films, almost all low budget explotation slasher pics. People such as D.W. Griffith, Mack Sennett, Alice Guy, Georges Méliès or Allan Dwan directed hundreds and hundreds of films, but most were 1 or 2 reelers and many did not officially credit them for their work.
Sennett alone has producer credit on over 1000 films.

The Guiness Book of Records gives the record to Indian director Dasari Narayana Rao with 151 films in Telugu, Tamil and Hindi. The exact number, however, seems to be disputed. Other sources state 148 or 150.
