Being from the UK and not knowing the first thing about baseball, I can only suggest going to: http://www.baseball-almanac.com/hitting/hi500c.shtml
which has the 500 club listing and puts Babe Ruth as the first.

Babe Ruth hit his 500th home run in 1929. Jimmie Foxx hit his 500th home run in 1940. They both were American League players at the time.
Mel Ott was the first National League player to reach the 500 home run milestone. He did so in 1945.
